Once you have reviewed the videos, prepare responses to the questions and discussion topics below, and solve the problems related to microbial identification. This will constitute your Laboratory

Template Report. There is no need to provide an introduction, general discussion or any other section. Submit your finalised Laboratory Template Report via the link with the same name at the bottom of the links under the heading “Assessment 3 – Lab Report” on the Moodle site.

Q1. Irradiation with UV light and wiping with ethanol are used to ensure a sterile working area in biological safety cabinets by killing microorganisms? How do these two methods accomplish this goal? 2 marks

Q2. Explain, using examples based on Covid-19 rapid antigen tests, what are false positive and false negative results. 2 marks

Q3. Why is there a much smaller number of antiviral drugs on the market compared to antibacterial drugs? 3 marks

Q4. During the video CHEM200 Combatting Microbes the results of an experiment in which E. coli was exposed to different antiseptics were presented. Which antiseptic was most effective? Justify your answer. 2 marks

Discussion topics

D1. The document “Disc Diffusion Experiment Results” shows 6 agar discs. Five of the agar discs contain one of the microbes from the list supplied, whilst the sixth contained none and gave an  invalid set of results. On each agar disc were placed 6 microdiscs containing different drugs. In addition, a seventh microdisc not containing any drug was also present. Analyse the results provided, and conduct additional research on the antibiotics and microorganisms, to identify which was the agar disc that gave the invalid set of results and which microorganism was present on each of the remaining agar discs. Provide explanations for your answers. 6 marks

D2. The document “API Strip Problems” shows the results obtained when microorganisms were subjected to different tests. These tests and the results they provide were explained in the video

CHEM200 Identifying Microbes. The results obtained from testing E. coli using the API strip that were presented during the above video are also shown in the API Strip Problems document. In addition, this document shows 3 other sets of results obtained using different microbes. Analyse these results using the API website to identify the 3 microbes. Also explain why this identification method does not always give a 100% match to a specific microbe. 5 marks
